Cholinium-based ionic liquids as bioinspired hydrotropes to tackle solubility challenges in drug formulation

Hydrotropy is a well-established strategy to enhance the aqueous solubility of hydrophobic drugs, facilitating their formulation for oral and dermal delivery. However, most hydrotropes studied so far possess toxicity issues are inefficient, with large amounts being needed achieve significant increases. Inspired by recent developments in understanding mechanism hydrotropy that reveal ionic liquids as powerful hydrotropes, present work use cholinium vanillate, gallate, salicylate two model ibuprofen naproxen, investigated. It shown vanillate gallate able increase up 500-fold, while all three revealed enhancements 600-fold case naproxen. Remarkably, increases 6000-fold. The results obtained exceptional hydrotropic ability cholinium-based even at diluted concentrations (below 1 mol·kg−1), when compared conventional hydrotropes. These especially relevant field drug due bio-based nature these low profiles. Finally, novel depend on synergism between both amphiphilic ions.
